Is it fine to use the PRV with any enclosure even the one has no locking cord?
Would be better to use the backfill adapter to vacuum the enclosure to make it more secure before the dive ? If yes what is the pressure should be on the gauge?

The PRV isn’t much use with an enclosure with no locking cord - if excess pressure builds up inside, the end cap is more likely to be pushed free of the enclosure rather than the pressure relief valve venting the extra gas.
The backfill adapter can be used to hold the enclosure shut with vacuum pressure - however if you pull a full vacuum the lack of air inside would make any electronics considerably more likely to overheat. Only a few psi of negative pressure (at most) is required to hold the enclosure shut - once the unit is more than a foot or two deep the water pressure makes the endcap very hard to remove…

So it is not beneficial to use it for the non locking cord enclosure. But it’s not an issue if I use the PRV instead of the enclosure vent ?

You can absolutely still use the PRV instead of the vent on a non locking enclosure, it will primarily serve only as a vent used when the enclosure is assembled rather than providing benefits as a pressure relief valve.
